(Negative)-Regulation of Cyclin-Dependent Kinases
The cell cycle is controlled by a series of checkpoints which when bypassed, enable the cell to proceed with division and replication. Cyclin-dependent kinases (CDKs) are the primary regulators of the cell cycle. CDKs are small with much of the peptide sequence representing its protein kinase domain; therefore, CDKs rely on interactions with cyclin regulatory subunits for enzyme activation and target specificity. Cyclins achieve temporal control of CDK function via fluctuations in their synthesis and degradation.
Deubiquitinases (DUBs)
play pivotal roles in the ubiquitin-proteasome cellular pathway. Due to their involvement in multiple key cell signaling processes
encompassing diseases such as cancer and neurodegeneration, these enzymes have evolved to be important new therapeutic targets.
